When you say you wouldn't change a thing about something, it means you are happy with the way it is now. It can be anything, coffee, hairdo or even life. It's perfect. You don't want to change anything, even if you can. Clooney is saying the coffee is perfect the way it is and it doesn't change even after spending a hard night.
