Welcome all to IS PHARMACOLOGY DIFFICULT Podcast ! I am Dr Radhika Vijay. In today's episode, I will be extending further my ongoing talk on Enzymes and today's ice breaking will e done by a word on Non equilibrium type of Enzyme Inhibition. It occurs with drugs which react with some catalytic site of the drug and reacts covalently or with such high affinity that the substrate is not able to react with the drug. It is more clarified with some great examples of Organophosphates or methotrexate . In both cases kM is unchanged, but Vmax is reduced. Then further, I will be giving a table of Non competitive inhibitors and their respective enzymes. So this will further make the whole concept crystal clear. Well, further giving a finishing touch to this whole talk, I will be talking that large variety of enzymes both intra and extra cellular, serve as molecular targets of drugs. Drugs may bind to active or allosteric site to produce the effect. This is again made clearer with an example of Angiotensin Converting Enzyme and Acetylcholine esterase. That was all I had to talk about Enzyme thing at present, discussion is infinite , but how to set limits to any talk in a relevant sensible way is surely an art of conversation.
