Precise DNA replication depends on the S-phase checkpoint and PCNA dynamics. Research shows PAF15 acts as a "toolbelt" anchor, coordinating Okazaki fragment processing by preventing steric clashes between enzymes. ATR inhibition causes excessive origin firing, exhausting resources.
