Nomhle started her acting career at the age of 19 years. In 2002, she received her Master’s degree in Theatre for Development in the United Kingdom from the King Alfred College.
Nkonyeni was the first black actress in theatre during the apartheid era in South Africa. She performed with different stage groups such as the Serpent Drama group.
