    eingesetzten Prostaglandinen kaum Nebenwirkungen.

Idiopathic epilepsy (IE) is a common breed-related neurological disorder in contemporary small animal medicine. The number of Border Collies (BCs) with epileptic seizures is increasing while there is a lack of data of IE in this breed. Hypothesis of this study was that IE occurs in BCs and manifests often with severe clinical signs and poor response to medical treatment.
IE was diagnosed by recurrent seizures, normal physical, neurological and laboratory examination. MRI and CSF analysis were requested if age at seizure onset was < 6 months or > 5 years of age. Dogs that failed to meet all inclusion criteria were only considered if a first- or second-degree relative was afflicted by IE or if seizures had occurred for at least 2 years without interictal neurological abnormalities. Owners fulfilled a detailed questionnaire. Subsequent phenotypic case classification was performed by evaluation of seizure history and treatment data. Pedigrees were sampled and matched for the appearance of common ancestors.
Ninety BCs with a reported seizure history were collected retrospectively and prospectively. Forty-nine of them were diagnosed with IE and were included in the present study. Clinical manifestations were dominated by moderate (33 %) and severe clinical courses (49 %) defined by the occurrence of cluster seizures or status epilepticus, respectively. Pharmacoresistance was apparent in 71 % of 24 dogs treated with ≥ 2 antiepileptic drugs. So far, no predictors for the occurrence of pharmacoresistance were identified in the present study. The epilepsy remission rate was 18 %. Dogs in remission showed a significantly higher median age at onset and a significantly lower initial seizure frequency compared to dogs with active epilepsy (p < 0.05). Survival time was significantly reduced in dogs aged < 2 years at seizure onset and in dogs with severe clinical courses (p < 0.05). Family- and pedigree analyses indicated a strong genetic founder effect in the appearance of epilepsy, resembling autosomal recessive inheritance. Yet, complex inheritance could not be excluded.
In conclusion, IE occurs in BCs and is frequently associated with severe clinical signs and pharmacoresistance. While further genetic research is required, the results of this study suggest a substantial hereditary (disease) component.

An important factor in the development and main¬tenance of neuropathic pain is a loss of glycinergic inhibition in the spinal cord. Here glycine is the major inhibitory neuro¬transmitter, but it also has excitatory functions as an obligate co-agonist of glutamate at the NMDA re¬ceptor. Glycine concentration in the synaptic cleft is controlled by glycine transporter 1 and glycine transporter 2. Inhibition of these two glycine transporters can influence glycinergic neuro¬transmission.
The goal of this study was to determine whether the intrathecal application of glycine transporter inhibitors ALX 5407 and ALX 1393, which would raise glycine levels in the synaptic cleft, can influence the nociception of rats. As gly¬cine acts on both the inhibitory glycine receptor and the excitatory NMDA receptor, we ex¬pected to see differential effects.
Bennett and Xie´s chronic constriction injury model was used as a model for neuro¬pathic pain. Hyperalgesia and allodynia develop ten to twelve days after loose ligation of the left sciatic nerve and can be measured to quantify neuropathic pain. An intrathe¬cal PE10 catheter was inserted into the vertebral canal to enable application of both gly¬cine transporter in¬hibitors (three dos¬ages respectively: 10, 50 and 100 μg dis¬solved in 10 μl 100 % DMSO; n = 8) in the vicinity of the dorsal horn of the spinal cord. Following the treatment, me¬chanical paw withdrawal threshold was measured using a von Frey-filament and thermal paw withdrawal latency was measured using a modified Har¬greaves method. Both measurements were made for 240 minutes.
The glycine transporter 1 inhibitor ALX 5407 had dose dependent differential effects on the noci¬cep¬tion of rats. Antinociceptive effects were obtained after application of the low and the high concentrations and pronociceptive effects were obtained after applica¬tion of the intermediate concentration. The pronociceptive actions were most prominent in the un¬injured right paw. There were no side effects. The glycine transporter 2 inhibitor ALX 1393 exerted exclusively antinociceptive ef¬fects that were significant only after appli¬cation of the high concentration. Additionally, at the high concentration, we observed severe neurological side effects in four of eight animals (pareses, paralysis, res¬piratory depres¬sion and analgesia).
The pronociceptive effects of glycine transporter 1 inhibitor ALX 5407 can be ex¬plained by an in¬creased activation of NMDA receptor through un¬hampered spill over of synaptically released glycine. The antinociceptive effects of both glycine transporter inhibitors and the neurological side effects of the glycine transporter 2 inhibitor ALX 1393 are likely to be mediated by the gly¬cine receptor.
This study shows, that neuropathic pain behaviour can be significantly in¬fluenced in vivo by manipulating glycine concentration through a singular intra¬the¬cal injection of spe¬cific glycine transporter inhibitors. The study of glycinergic neurotransmission with the aid of glycine transporter inhibi¬tors promises future development of potent and versatile therapeutics. These may have potential to treat neuropathic pain, act as muscle relaxants and analgesics and also function as cognitive enhancers and treatments for schizo¬phrenia.

  • Noninvasive assessment of retinal morphology in mice using optical coherence tomography
Animal models are important organisms in many areas of science. They play a key role in experimental ophthalmology because they help to understand a variety of genetical, developmental, and disease mechanisms and to develop new pharmaceutical and gene therapies. Especially mice are valuable models to identify the genes involved in vision because of the availability of diverse genetically modified strains and the ease with which single gene mutants can be generated.
The retina as part of the brain offers the opportunity to directly visualize changes associated with neurodegenerative disorders and vascular alterations. There are both morphological and functional approaches to characterize disease phenotypes, to monitor disease progression, and to evaluate the responsiveness to therapy, which can either be performed in living animals (in vivo) or in respective ocular tissue (in vitro).
Whereas most functional tests, namely electroretinography (ERG), are performed in vivo, practically all morphological methods, like histology, are so far performed in vitro. The current need to sacrifice animals for histological examinations at different time points interferes with the ability to follow up disease processes and to monitor therapeutic or side effects during the preclinical assessment of novel genetical and pharmaceutical therapy strategies over time in the same individuals.
Optical coherence tomography (OCT) is a novel technique to assess retinal morphology in vivo. Commercially available OCTs have been designed for clinical investigations in human ophthalmology. In this work, the establishment of a commercially available OCT for the in vivo analysis of mouse models of retinal degenerations is reported.
