The ark represented and symbolised the presence of God with Israel.

If our hearts are obedient and open, if we are striving to glorify God in our lives and we are truly guided by His word on this then the glory will not depart. But if we get opinionated, disobedient, independent, immoral then God will remove his glory and blessing from us as he did from Israel at this time.
