Neurofibromatosis type 1 (NF1) is an incurable and progressive genetic disorder caused by a partial or complete loss of function of the NF1 tumor suppressor gene, resulting in elevated RAS-mitogen-activated protein kinase (RAS/MAPK) pathway signaling and the potential development of subsequent NF1-associated tumors. Given current breakthroughs, including the emergence of MEK inhibitors, the ongoing challenge is how to effectively employ modern therapeutics in the management of patients with NF1 tumors, particularly pediatric patients.
