Recently approved immunotherapy options have revolutionized the treatment of patients with advanced gastroesophageal cancers (eg, gastric adenocarcinoma, gastroesophageal junction adenocarcinoma, esophageal adenocarcinoma, esophageal squamous cell carcinoma [ESCC]), which have traditionally been associated with poor outcomes due to late detection, high heterogeneity, and limited response to chemotherapy and targeted treatments. With clinical trials validating the efficacy of immune checkpoint inhibition across multiple lines of therapy and regardless of PD-L1 expression, nivolumab and pembrolizumab have become standards of care in the frontline setting as part of various combinations, and nivolumab monotherapy has been approved in patients with ESCC after previous chemotherapy. Recently, nivolumab plus ipilimumab became the first chemotherapy-free combination to gain approval for frontline use in patients with ESCC.
